Sharon Jones and the Dap-Kings recorded 100 Days, 100 Nights at Daptone's House of Soul in Brooklyn, using the label's analogue studio setup. Bosco Mann's production leaves the horns, rhythm section and Jones's voice close enough together to sound like a band responding in the moment.

The title song slows and changes shape as Jones weighs a lover's intentions. “When the Other Foot Drops, Uncle” has a more forceful attack, and “Answer Me” draws on gospel. The performances give the album movement within its deliberately compact soul arrangements.
